I have changed a dtd used by a configured op on xml adapter. I need to know if I need restart the adapter to relaod the changed dtd or it will be automatically loaded into the adapter?

Yes, you will definitely need to restart the adapter before you see the changes. When the adapter starts, it initializes any configured operations. When it does this, it gets the current DTD or XML schema in the defined directory. I believe it holds this in memory. So if you place a new DTD in that directory, it will not recognize it until the adapter is restarted and the operation is re-initialized.

You don’t need to with XML Adapter 4.2. Just hit the refresh arrow. The Best thing to do though is not to completely replace the DTD but just edit the changes so you won’t have to remapp everything.
